Localization of Carboxypeptidase I in Germinating Barley Grain 1
نویسندگان
چکیده
Activity measurements and Northem blot hybridizations were used to study the temporal and spatial expression of carboxypeptidase I in germinating grains of barley (Hordeum vulgare L. cv Himalaya). In the resting grain no carboxypeptidase I activity was found in the aleurone layer, scutellum, or starchy endosperm. During germination high levels of enzyme activity appeared in the scutellum and in the starchy endosperm but only low activity was found in the aleurone layer. No mRNA for carboxypeptidase I was observed in the resting grain. By day 1 of germination the mRNA appeared in the scutellum where its level remained high for several days. In contrast, little mRNA was observed in the aleurone layer. These results indicate that the scutellum plays an important role in the production of carboxypeptidase I in germinating barley grain. or to secretion from the surrounding living tissues, the aleurone layer, or the scutellum. The aim of the present study is to determine whether carboxypeptidase I is synthesized during germination and if so-whether the synthesis takes place in the scutellum or in the aleurone layer. In addition to enzyme activity, mRNA for this enzyme was also localized. MATERIALS AND METHODS

متن کاملAcid Carboxypeptidases in Grains and Leaves of Wheat, Triticum aestivum L.
Extracts of resting and germinating (3 days at 20 degrees C) wheat (Triticum aestivum L. cv Ruso) grains rapidly hydrolyzed various benzyloxycarbonyldipeptides (Z-dipeptides) at pH 4 to 6. Similar activities were present in extracts of mature flag leaves. Fractionation by chromatography on CM-cellulose and on Sephadex G-200 showed that the activities in germinating grains were due to five acid ...
متن کاملDetermination of Endogenous Gibberellins in Germinating Barley by Combined Gas Chromatography-Mass Spectrometry
This study was undertaken to elucidate the dominant endogenous gibberellins (GA) and the changes in their levels in germinating barley during malting. Also, a method for determining gibberellins in barley and malt by combined gas chromatography-mass spectrometry (GC-MS) was developed. Endogenous gibberellins in germinating Fuji Nijo I I and Betzes barley were sequentially extracted with ethanol...

In cereals, senescence is a highly regulated and organized process in which the nutrients located in the leaves are translocated to the developing seeds. During this process multiple signaling cascades are activated either up-regulating or down-regulating senescence related genes. The majority of the nitrogen to be remobilized is located in the leaves in plastidial proteins, especially Rubisco....
متن کاملBeta-amylase degradation by serine endoproteinases from green barley malt
Proteolytic degradation of barley proteins is examined in green (unkilned) malt and germinating seeds from Hordeum vulgare L. cv. Harrington. Zymographic analysis of the Harrington green malt extracts using commercial preparations of barley beta-amylase incorporated as a proteolytic substrate in 2-D SDS gels shows multiple proteolytic activities. A developmental study shows that the several gre...
